چکیده

Production of manganese-dependent peroxidase (MnP) by the white-rot fungus Phanerochaete chrysosporium BKMF-1767 (ATCC 24725) was monitored during growth in different media and growth conditions. The effect of some activators of MnP production, Mn, Tween 80, phenylmethylsulphonylfloride (PMSF), oxygen, temperature, pH, glycerol and nitrogen was studied. Supplementing the cultures with Tween 80 (0.05 %, v/v) and Mn (174 μM) resulted a maximum MnP activity of 356 U/L which was approximately two times higher than that obtained in the control culture (without Tween 80). Decolourisation of Direct Blue 15 and Direct Green 6 (50 mg/L) was also

Lipid Peroxidation by the Manganese Peroxidase of Phanerochaete chrysosporium Is the Basis for Phenanthrene Oxidation

The manganese peroxidase (MnP) of Phanerochaete chrysosporium supported Mn(II)-dependent, H202independent lipid peroxidation, as shown by two findings: linolenic acid was peroxidized to give products that reacted with thiobarbituric acid, and linoleic acid was peroxidized to give hexanal. MnP also supported the slow oxidation of phenanthrene to 2,2'-diphenic acid in a reaction that required Mn(...

HYPERPRODUCTIVITY OF EXTRACELLULAR ENZYMES FROM INDIGENOUS WHITE ROT FUNGI (P. chrysosporium IBL-03) BY UTILIZING AGRO-WASTES

An indigenous locally isolated white rot fungal strain Phanerochaete chrysosporium IBL-03 was investigated for the hyper-production of ligninolytic enzymes from different agro-industrial wastes including wheat straw, rice straw, banana stalks, corncobs, corn stover, and sugarcane bagasse as substrate material in still culture fermentation technique. Screening experiments were performed at 30C f...

Biodegradation of 2,4-dichlorophenol in a fluidized bed reactor with immobilized Phanerochaete chrysosporium.

The performance of a fluidized bed reactor using immobilized Phanerochaete chrysosporium to remove 2,4-dichlorophenol (2,4-DCP) from aqueous solution was investigated. The contribution of lignin peroxidase (LiP) and manganese peroxidase (MnP) secreted by Phanerochaete chrysosporium to the 2,4-DCP degradation was examined. Results showed that Lip and Mnp were not essential to 2,4-DCP degradation...

Novel promoter sequence required for manganese regulation of manganese peroxidase isozyme 1 gene expression in Phanerochaete chrysosporium.

Manganese peroxidase (MnP) is a major, extracellular component of the lignin-degrading system produced by the wood-rotting basidiomycetous fungus Phanerochaete chrysosporium. The transcription of MnP-encoding genes (mnps) in P. chrysosporium occurs as a secondary metabolic event, triggered by nutrient-nitrogen limitation. In addition, mnp expression occurs only under Mn2+ supplementation. Using...

Roles of manganese and organic acid chelators in regulating lignin degradation and biosynthesis of peroxidases by Phanerochaete chrysosporium.

We studied the effect of manganese and various organic chelators on the distribution, depolymerization, and mineralization of synthetic 14C-labeled lignins (DHP) in cultures of Phanerochaete chrysosporium. In the presence of high levels of manganese [Mn(II) or Mn(III)], along with a suitable chelator, lignin peroxidase (LiP) production was repressed and manganese peroxidase (MnP) production was...
